Der Verlust der Biodiversität wird sowohl auf einer globalen Skala als auch für die anthropogen geformten Landschaften, die heute fast 50% der terrestrischen Landfläche ausmachen, festgestellt. Auf den landwirtschaftlichen Anbauflächen werden Pestizide, biologisch aktive Chemikalien, ausgebracht um Schädlinge, Krankheiten und Unkräuter zu kontrollieren. Um die Auswirkung der Pestizide auf die Biodiversität zu verstehen ist die Quantifizierung der verbliebenen semi-natürlichen Strukturen wie Feldsäume und Hecken, die Organismen in Agrarlandschaften als Habitat dienen, eine Voraussetzung. Für eine Abschätzung ihrer potentiellen Pestizidexposition ist zudem die Anwesenheit der Organismen in diesen Habitaten und in den Feldkulturen notwendig. Im vorliegenden Text stelle ich Studien für Tiergruppen wie Amphibien, Fledermäuse und Motten vor, die bisher nicht in der Risikobewertung für Pestizide berücksichtigt worden sind. Für alle Gruppen wurde dargelegt, dass sie sowohl in der Agrarlandschaft leben als auch potentiell mit Pestiziden in Kontakt kommen und daher ein Risiko angezeigt ist. Für die Risikobetrachtung sind auch Informationen zur Empfindlichkeit der Organismen notwendig und hier werden neue Daten für Pflanzen, Amphibien und Bienen vorgestellt. Effekte die bis auf die Gemeinschaftsebene wirksam waren, wurden für die Auswirkungen von Herbizid, Insektizid und Dünger in einem natürlichen System betrachtet. Das Ergebnis nach drei Behandlungsjahren waren vereinfachte Pflanzengemeinschaften mit geringerer Artenzahl und einer reduzierten Anzahl von Blütenpflanzen. Die Abnahme an Blüten stellt ein Beispiel eines indirekten Effekts dar und war für die Effekte eines Herbizids auf den scharfen Hahnenfuß besonders auffällig. Subletale Herbizideffekte für Pflanzen hatten einen Einfluss auf daran fressende Raupen was durch eine Verminderung der Nahrungsqualität erklärbar ist. Für Feldsäume realistische Insektizidmengen reduzierten die Bestäubung der weißen Lichtnelke durch Motten um 30%. Diese indirekten Effekte durch Veränderungen im Nahrungsnetz spielen eine kritische Rolle für das Verständnis des Rückgangs von verschiedenen Organismengruppen, allerdings werden sie bisher nicht in die Risikobewertung von Pestiziden mit einbezogen. Der aktuelle intensive Pestizideinsatz in der Landwirtschaft und ihre hohe Toxizität könnten zu einer chemisch fragmentierten Landschaft führen in der Population nicht mehr verbunden sind und damit deren Größe und genetische Struktur beeinflussen. Die Modellierung von möglichen Pestizideffekten als Kosten für die Anwanderung von Amphibien zu Fortpflanzungsgewässern in Weinbergen in Rheinland-Pfalz zeigte die Isolation der untersuchten Populationen an. Eine erste Validierung für den Grasfrosch bestätigte die Modellvorhersagen für einige Populationen. Für den terrestrischen Bereich der Risiko-bewertung ist eine Vielzahl von Richtlinien vorhanden oder wird aktuell entwickelt oder verbessert. Die Ergebnisse der vorliegenden Arbeiten zeigen, dass vor allem die reproduktiven Blütenstadien von Pflanzen sehr empfindlich sind und ihr Risiko unterschätz ist. Die Erholung von Arthropodenpopulationen nach Pestizideffekten muss auf Landschafts-ebene neu bemessen werden und eine Risikobewertung für Amphibien für die Zulassung wird vorgeschlagen. Die Etablierung und Anpassung von Risikobewertungssystemen ist allerdings ein zeitaufwändiger Prozess und daher stellt die Entwicklung von Risikomanagementmaßnahmen eine pragmatische Alternative mit unmittelbaren Auswirkungen dar. Künstliche Gewässer der Agrarlandschaft sind wichtige Nahrungsgebiete für Fledermäuse und ihre Anlage würde negative Auswirkungen des Pestizideinsatzes abschwächen. Die Einbindung von direkten und indirekten Effekten für alle Organismengruppen in eine Risikobewertung in der auch der Landschaftsmaßstab und Pestizidmischungen betrachtet werden wird viel Entwicklungszeit benötigen. Die Etablierung von Modellandschaften in der Managementmaßnahmen und integrierter Pflanzenschutz auf größerer Skala angewendet werden, würde es uns jedoch erlauben die Auswirkungen von Pestiziden in einem realistischen Szenario zu untersuchen und Ansätze für die Landwirtschaft der Zukunft zu entwickeln.
With 47% land coverage in 2016, agricultural land was one of the largest terrestrial biomes in Germany. About 70% of the agricultural land was cropped area with associated pesticide applications. Agricultural land also represents an essential habitat for amphibians. Therefore, exposure of amphibians to agrochemicals, such as fertilizers and pesticides, seems likely. Pesticides can be highly toxic for amphibians, even a fraction of the original application rate may result in high amphibian mortality.
To evaluate the potential risk of pesticide exposure for amphibians, the temporal coincidence of amphibian presence on agricultural land and pesticide applications (N = 331) was analyzed for the fire-bellied toad (Bombina bombina), moor frog (Rana arvalis), spadefoot toad (Pelobates fuscus) and crested newt (Triturus cristatus) during spring migration. In 2007 and 2008, up to 80% of the migrating amphibians temporally coincided with pesticide applications in the study area of Müncheberg, about 50 km east of Berlin. Pesticide interception by plants ranged between 50 to 90% in winter cereals and 80 to 90% in winter rape. The highest coincidence was observed for the spadefoot toad, where 86.6% of the reproducing population was affected by a single pesticide in winter rape during stem elongation with 80% pesticide interception by plants. Late migrating species, such as the fire-bellied toad and the spadefoot toad, overlapped more with pesticide applications than early migrating species, such as the moor frog, did. Under favorable circumstances, the majority of early migrants may not coincide with the pesticide applications of arable fields during spring migration.
To evaluate the potential effect of pesticide applications on populations of the common frog (Rana temporaria), a landscape genetic study was conducted in the vinicultural area of Southern Palatinate. Due to small sample sizes at breeding sites within viniculture, several DNA sampling methods were tested. Furthermore, the novel repeated randomized selection of genotypes approach was developed to utilize genetic data from siblings for more reliable estimates of genetic parameters. Genetic analyses highlighted three of the breeding site populations located in viniculture as isolated from the meta-population. Genetic differentiation among breeding site populations in the viniculture (median pairwise FST=0.0215 at 2.34 km to 0.0987 at 2.39 km distance) was higher compared to genetic differentiation among breeding site populations in the Palatinate Forest (median pairwise FST=0.0041 at 5.39 km to 0.0159 at 9.40 km distance).
The presented studies add valuable information about the risk of pesticide exposure for amphibians in the terrestrial life stage and possible effects of agricultural land on amphibian meta-populations. To conserve endemic amphibian species and their (genetic) diversity in the long run, the risk assessment of pesticides and applied agricultural management measures need to be adjusted to protect amphibians adequately. In addition, other conservation measures such as the creation of new suitable breeding site should be considered to improve connectivity between breeding site populations and ensure the persistence of amphibians in the agricultural land.
Factors triggering the ecotoxicity of metal-based nanoparticles towards aquatic invertebrates

Heutzutage werden Nanopartikel in großem Maßstab produziert, weshalb deren Eintrag in Oberflächengewässer immer wahrscheinlicher wird. Dort angelangt unterliegen sie verschiedenen umweltbedingten (Oberflächen-)Modifikationen, die in letzter Konsequenz eine Vielfalt von Nanopartikel-Agglomeraten unterschiedlicher Größe hervorbringen. Direkt davon betroffen sind aquatische Lebewesen, die einer entsprechenden Nanopartikelexposition in der Wasserphase ausgesetzt sind.
Nach Sedimentation der Agglomerate können aber ebenfalls benthische Organismen betroffen sein. Bisherige ökotoxikologische Untersuchungen haben solche umweltbedingten Einflüsse außer Acht gelassen und viel mehr nanopartikel-spezifische Charakteristika auf deren Wirkweise gegenüber pelagischen Vertretern untersucht. Aus diesem Grund ist eine systematische Untersuchung derer Faktoren von Nöten, die den Verbleib und das Verhalten aber auch die Toxizität von Nanopartikeln in der Umwelt maßgeblich beeinflussen. Die kumulative Arbeit dieser Dissertation macht sich dies zum Ziel und hinterfragt entsprechende Faktoren die einerseits durch Nanopartikel assoziierte Aspekte (definiert als i) inhärente Stoffeigenschaft des untersuchten Materials und ii) Nanopartikel Charakteristika)) und andererseits durch Umweltbedingungen in Oberflächengewässern geprägt sind. In diesem Kontext wurden verschiedene ökotoxikologische Untersuchungen mit inerten Titandioxid Nanopartikeln (nTiO2) und Ionen freisetzenden Silber Nanopartikeln (nAg) unter Berücksichtigung verschiedener Nanopartikel Charakteristika (z.B. initiale Partikelgröße, Oberflächengröße) und Umweltbedingungen (z.B. Ionenstärke, ultraviolettes Licht (UV-Licht)), durchgeführt.
Als Testorganismen dienten dazu die pelagischen bzw. benthischen Vertreter Daphnia magna und Gammarus fossarum. Die Ergebnisse deuten daraufhin, dass die Toxizität von nTiO2 und nAg gegenüber Daphnien maßgeblich durch das Adsorptionspotential (im Bezug auf das Anhaften der Partikel an die Organismenoberfläche) und das Umweltverhalten (Freisetzung von radikalen Sauerstoffspezies oder Metallionen) der Nanopartikel bestimmt wird.
Darüber hinaus wurde die Nanopartikeltoxizität von jenen inhärenten Stoffeigenschaften, Nanopartikelcharakteritika und Umweltbedingungen am meisten beeinflusst, welche die zuvor genannten Aspekte entweder verstärken oder abschwächen. Hierfür beispielhaft ist der toxizitätsverstärkende Effekt von UV-Licht auf nTiO2 in Experimenten mit Gammarus: Während eine Exposition der Organismen in absoluter Dunkelheit selbst bei 5,00 mg nTiO2/L keine Effekt hervorrief, kam es in der Anwesenheit von UV-Licht schon bei 0,20 mg nTiO2/L zu schwerwiegenden Effekten auf sublethaler und lethaler Ebene.
Unter Berücksichtigung der Ergebnisse dieser Dissertation sowie bisherige Erkenntnisse der Wissenschaft im Allgemeinen, ist die derzeitige Risikoeinschätzung von Nanopartikeln möglicherweise unprotektiv, sofern eine Interaktion von Nanopartikeln und Umwelteinflüssen unberücksichtigt bleibt
The European weatherfish (Misgurnus fossilis) is a benthic freshwater fish species belonging to the family Cobitidae, that is subjected to a considerable decline in many regions across its original distribution range. Due to its cryptic behavior and low economic value, the causes of threat to weatherfish remained partly unknown and the species is rarely at the center of conservation efforts. In order to address these concerns, the overall aim of the present thesis was to provide a comprehensive approach for weatherfish conservation, including the development of stocking measures, investigations on the species autecology and the evaluation of potential threats. The first objective was to devise and implement a regional reintroduction and stock enhancement program with hatchery-reared weatherfish in Germany. Within this program (2014-2016), a total number of 168,500 juvenile weatherfish were stocked to seven water systems. Recaptures of 45 individuals at two reintroduction sites supported the conclusion that the developed stocking strategy was appropriate. In order to broaden the knowledge about weatherfish autecology and thereby refining the rearing conditions and the selection of appropriate stocking waters, the second objective was to investigate the thermal requirements of weatherfish larvae. Here, the obtained results revealed that temperatures higher than previously suggested were tolerated by larvae, whereas low temperatures within the range of likely habitat conditions increased mortality rates. As weatherfish can be frequently found in agriculturally impacted waters (e.g. ditch systems), they are assumed to have an increased probability to be exposed to chemical stress. Since the resulting risk has not yet been investigated with a focus on weatherfish, the third objective was to provide a methodical foundation for toxicity testing that additionally complies with the requirements of alternative test methods. For this purpose, the acute fish embryo toxicity test was successfully transferred to weatherfish and first results exhibited that sensitivity of weatherfish towards a tested reference substance (3,4-dichloroaniline) was highest compared to other species. On the basis of these findings, the fourth objective was to apply weatherfish embryos for multiple sediment bioassays in order to investigate teratogenic effects derived from sediment-associated contaminants. In this context, weatherfish revealed particular sensitivity to water extractable substances, indicating that sediment contamination might pose a considerable risk. Moreover, as an endangered benthic fish species with high ecological relevance for European waters that are specifically exposed to hazardous contaminants, the weatherfish might be a prospective species for an ecological risk assessment of sediment toxicity. Overall, the present thesis contributed to the conservation of weatherfish by considering a variety of aspects that interact and reinforce one another in order to achieve improvements for the species situation.
In einem Großteil der Welt wird Grundwasser für die Versorgung von Siedlungen und Agrarflächen genutzt. Organismen, die im Grundwasser leben, erfüllen wichtige Funktionen im Ökosystem und haben positiven Einfluss auf die Grundwasserqualität. Um das Risiko negativer Effekte auf diese wertvollen Ökosysteme zu minimieren muss die entsprechende Sanierungsmethode, im Falle einer Grundwasserbehandlung, mit Vorsicht gewählt werden. In der vorliegenden Thesis wurde das Umweltrisiko von Carbo-Iron untersucht, ein Komposit aus nanoskaligem null-valentem Eisen und Aktivkohle zur in situ-Behandlung von Grundwasser. Des Weiteren wurde eine umfassende Beurteilung des Umweltrisikos und des Nutzens einer Grundwasserbehandlung mit Carbo-Iron durchgeführt.
Zu Beginn der Arbeit an der vorliegenden Thesis existierten noch keine Empfehlungen für Untersuchung der Ökotoxizität von Nanomaterialien. Daher bestanden viele Unsicherheiten hinsichtlich geeigneter Methoden. Im Rahmen dieser Thesis wurde eine Entscheidungshilfe entwickelt, um bei der ökotoxikologischen Untersuchung von Nanomaterialien systematisch geeignete methodische Schritte auszuwählen.
Mögliche Effekte von Carbo-Iron wurden in Tests mit embryonalen, juvenilen und adulten Lebensstadien des Zebrabärblings (Danio rerio) und juvenilen und adulten Amphipoden (Hyalella azteca) untersucht. Die gewählten Testsysteme basierten auf existierenden Testmethoden der OECD und EPA zur ökotoxikologischen Untersuchung von Chemikalien (OECD, 1992a, 2013a, 2013b; US EPA, 2000). Zusätzlich wurde die Aufnahme der Partikel in die genannten Testorganismen untersucht. In Zebrabärblingsembryonen wurden außerdem potentielle Effekte auf die Genexpression mittels Microarrays ermittelt. Die erhaltenen Daten wurden später mit Ergebnissen aus Tests mit dem Wasserfloh Daphnia magna, der Alge Scenedesmus vacuolatus, Larven der Mücke Chironomus riparius und nitrifizierenden Bodenmikroorganismen ergänzt.
In dem Fischembryotoxizitätstest wurde keine Passage der Carbo-Iron-Partikel durch das Chorion in den perivitellinen Raum oder den Embryo beobachtet. Nach der Exposition wurde Carbo-Iron im Darm von H. azteca und D. rerio, aber keinem anderen Gewebe oder Organen detektiert. Carbo-Iron hatte keine signifikanten Effekte auf die Nitrifikationsrate der Bodenmikroorganismen sowie Überleben und Wachstum des Zebrabärblings. Dennoch wurden signifikant negative Effekte auf Wachstum, Fütterungsrate und Reproduktion von H. azteca und auf das Überleben und die Reproduktion von D. magna festgestellt. Des Weiteren war die Entwicklungsrate von C. riparius und das Zellvolumen von S. vacuolatus negativ beeinflusst.
Anhand der durchgeführten Studien wurde basierend auf dem Ergebnis des Reproduktionstests mit D. magna und einem assessment factor von 10 für Carbo-Iron eine predicted no effect concentration von 0,1 mg/L ermittelt. Diese wurde mit modellierten und gemessenen Umweltkonzentrationen von Carbo-Iron verglichen die in einer Studie erhoben wurden, in denen Carbo-Iron zur Behandlung eines mit Chlorkohlenwasserstoffen kontaminierten Aquifers eingesetzt wurde, und Risiko-Quotienten wurden abgeleitet. Zur gesamtheitlichen Betrachtung wurde anschließend ein Schema zur Bewertung des Umweltrisikos vor und nach der Behandlung des Aquifers mit Carbo-Iron entwickelt. Die erhobenen Daten weisen auf ein reduziertes Umweltrisiko nach der Applikation von Carbo-Iron hin. Dementsprechend überwiegen die Vorteile einer Grundwasserbehandlung mit Carbo-Iron die potentiellen negativen Effekte auf die Umwelt.
Studies have shown that runoff and spray-drift are important sources of nonpoint-source pesticide pollution of surface waters. Owing to this, public concern over the presence of pesticides in surface and ground water has resulted in intensive scientific efforts to find economical, yet environmentally sound solutions to the problem. The primary objective of this research was to assess the effectiveness of vegetated aquatic systems in providing buffering between natural aquatic ecosystems and agricultural landscape following insecticide associated runoff and spray-drift events. The first set of studies were implemented using vegetated agricultural ditches, one in Mississippi, USA, using pyrethroids (bifenthrin, lambda-cyhalothrin) under simulated runoff conditions and the other in the Western Cape, South Africa using the organophosphate insecticide, azinphos-methyl (AZP), under natural runoff and spray-drift conditions. The second set of studies were implemented using constructed wetlands, one in the Western Cape using AZP under natural spray-drift conditions and the other in Mississippi, USA using the organophosphate MeP under simulated runoff conditions. Results from the Mississippi-ditch study indicated that ditch lengths of less than 300 m would be sufficient to mitigate bifenthrin and lambda-cyhalothrin. In addition, data from mass balance calculations determined that the ditch plants were the major sink (generally > 90%) and/or sorption site for the rapid dissipation of the above pyrethroids from the water column. Similarly, results from the ditch study in South Africa showed that a 180 m vegetated system was effective in mitigating AZP after natural spray drift and low flow runoff events. Analytical results from the first wetland study show that the vegetated wetland was more effective than the non-vegetated wetland in reducing loadings of MeP. Mass balance calculations indicated approximately 90% of MeP mass was associated with the plant compartment. Ninety-six hours after the contamination, a significant negative acute effect of contamination on abundances was found in 8 out of the 15 macroinvertebrate species in both wetland systems. Even with these toxic effects, the overall reaction of macroinvertebrates clearly demonstrated that the impact of MeP in the vegetated wetland was considerably lower than in the non-vegetated wetland. Results from the constructed wetland study in South Africa revealed that concentrations of AZP at the inlet of the 134 m wetland system were reduced by 90% at the outlet. Overall, results from all of the studies in this thesis indicate that the presence of the plant compartment was essential for the effective mitigation of insecticide contamination introduced after both simulated and natural runoff or spray-drift events. Finally, both the vegetated agricultural drainage ditch and vegetated constructed wetland systems studied would be effective in mitigating pesticide loadings introduced from either runoff or spray-drift, in turn lowering or eliminating potential pesticide associated toxic effects in receiving aquatic ecosystems. Data produced in this research provide important information to reduce insecticide risk in exposure assessment scenarios. It should be noted that incorporating these types of best management practices (BMPs) will decrease the risk of acute toxicity, but chronic exposure may still be an apparent overall risk.
Die Abschätzung der Eintragswahrscheinlichkeit von Pestiziden in Gewässer ist insbesondere im Landschaftsmaßstab aufgrund der erforderlichen ereignisbezogenen Probenahmetechniken mit erheblichem Kostenaufwand verbunden. Simulationsmodelle können hierbei eine wichtige Alternative zur Planung von Risiko-Minimierungsmaßnahmen darstellen. Hauptziele dieser Arbeit waren (A) die Entwicklung eines Simulationswerkzeugs, mit dessen Hilfe das Eintragsrisiko von Pestiziden in Oberflächengewässer im Landschaftsmaßstab abgeschätzt werden kann und sich Risiko-Minimierungsmaßnahmen effektiv planen lassen, sowie (B) die Durchführung einer Expositions- und Risikoabschätzung sowie die Abschätzung negativer Effekte von Pestiziden auf die Artenzahlen von Fließgewässer-Lebensgemeinschaften des Braunschweiger Umlands. Exposition, Risiko und Effekte In den Abschnitten 1.1 und 1.2 wurde das Simulationsmodell auf landwirtschaftlich beeinflusste Fließgewässer des Braunschweiger Umlands angewendet. Abschnitt 1.3 gibt einen Überblick über allgemeine Anwendungsmöglichkeiten des Simulators und den Systemaufbau. Abschnitt 1.1 - Scenario based simulation of runoff-related pesticide entries into small streams on a landscape level (Englische Publikation, S. 27): Auf Basis des von der OECD vorgeschlagenen (ratio of exposure to toxicity) REXTOX-Modells wurde der Pestizideintrag für 737 Probestellen bei kleineren Fließgewässern des Braunschweiger Umlands simuliert. Die Simulation wurde für die 15 im Untersuchungsgebiet am häufigsten nachgewiesenen Pestizide für je acht verschiedene Umweltszenarien durchgeführt. Die Szenarien umfassten Management-Maßnahmen, wie z.B. die Variation der Randstreifenbreite, sowie klimatische und saisonale Aspekte. Die höchsten Konzentrationen wurden für ein Szenario ohne Randstreifen und gleichzeitig erhöhten Niederschlägen vorhergesagt. Anhand der simulierten Konzentrationen wurde das Risiko für aquatische Lebensgemeinschaften auf Basis von Standard-Toxizitätstests (Endpunkte EC50, LC50) und eines Sicherheitsfaktors abgeschätzt. Abschnitt 1.2 - Linking land use variables and invertebrate taxon richness in small and medium-sized agricultural streams on a landscape level (Englische Publikation, S. 50): In dieser Studie wurden die langjährigen mittleren Artenzahlen von Fließgewässer-Invertebratentaxa im landwirtschaftlich geprägten Braunschweiger Umland (Untersuchungen über 15 Jahre bei 90 Gewässern an 202 Probestellen) auf Wechselbeziehungen mit verschiedenen Umweltfaktoren, wie z.B. der Gewässerbreite, der Landnutzung (Landwirtschaft, Wald, Wiese, Siedlung), der Bodenart sowie landwirtschaftlich bedingter Stressoren untersucht. Der Stress, der von der landwirtschaftlichen Nutzung ausgeht, wurde dabei durch das Risiko des Auftretens von Oberflächenabfluss (runoff) von der Ackerfläche in das Gewässer ausgedrückt. Der Faktor "Risk of Runoff" wurde von dem von der OECD vorgeschlagenen "ratio of exposure to toxicity" (REXTOX)-Modell abgeleitet. Durch multivariate Statistik konnten insgesamt 39,9% der Varianz in der Artenzahl erklärt werden, wobei die Gewässerbreite mit 25,3% den wichtigsten Faktor darstellte, gefolgt vom Faktor "Risk of Runoff" mit 9,7%. Abschnitt 1.3 - Informationssystem zur ökotoxikologischen Bewertung der Gewässergüte in Bezug auf Pflanzenschutzmitteleinträge aus der Landwirtschaft - Systemaufbau und Anwendungsmöglichkeiten (Tagungsbeitrag S. 61): Abschnitt 1.3 enthält einen kurzen Überblick über das Simulationssystem, generelle Anwendungsmöglichkeiten sowie Anwendungsbeispiele. Das Simulationssystem Ein wichtiger Aspekt für den Einsatz eines Simulationsmodells im Rahmen der Risikoabschätzung ist dessen einfache Anwendbarkeit in der Praxis. Dazu zählen die leichte Verfügbarkeit der benötigten Eingangsdaten, die Umsetzung des Modells in eine Softwareanwendung sowie die entsprechende Dokumentation des Systems. Abschnitt 1.4 - Informationssystem zur ökotoxikologischen Bewertung der Gewässergüte in Bezug auf Pflanzenschutzmitteleinträge aus der Landwirtschaft - Simulationsmodell und Systemaufbau (BMBF-Bericht, S. 67): In diesem Abschnitt wird das verwendete Modell, die "simplified formula for indirect loadings caused by runoff" (SFIL), der schematische Aufbau des Simulationssystems sowie der Ablauf des Datenverarbeitungsprozesses dargestellt. Abschnitt 1.5 - Benutzerhandbuch (BMBF-Bericht, S. 71): Das Benutzerhandbuch enthält Informationen über die Installation des Systems, die Erzeugung der benötigten Eingangsdaten, zur allgemeinen Arbeit mit dem System sowie Anwendungsbeispiele (Was-wäre-wenn-Analyse). Abschnitt 1.6 - Technische Dokumentation (BMBF-Bericht, S. 104): Die technische Dokumentation beschreibt die Struktur der Import-/Export-Datenbanktabellen des Simulators.
Etwa 50 % der Fläche Europas werden landwirtschaftlich genutzt. Dennoch gibt es nahezu keine Information ob Fledermäuse diese Flächen beispielsweise zur Nahrungsaufnahme nutzen. Aufgrund der limitierten Datenbasis mangelt es auch an Schutzkonzepten, die mögliche negative Effekte der landwirtschaftlichen Intensivierung auf die Fledermäuse und deren Nahrungsgrundlage ausgleichen könnten. Da die Exposition von Fledermäusen mit Pflanzenschutzmitteln bislang nicht thematisiert wurde, sind im europäischen Zulassungsverfahren für Pflanzenschutzmittel keine Risikoabschätzungen für Fledermäuse gefordert. Um für Fledermäuse die mögliche Exposition gegenüber Pflanzenschutzmitteln abschätzen zu können, sind Informationen über Vorkommen und Aktivität von Fledermäusen in landwirtschaftlich genutzten Flächen erforderlich. Die Erfassung von Fledermäusen auf einer Vielzahl von Flächen machte es notwendig sich im Vorfeld auf eine geeignete Methodik festzulegen. Die akustische Fledermauserfassung ist im Gegensatz zu deutlich zeitaufwändigeren Methoden wie Netzfang, Telemetrie oder direktes Beobachten die einzige logistisch durchführbare Methode. In der wissenschaftlichen Literatur werden jedoch bei vielen bisher durchgeführten akustischen Methoden Bedenken bezüglich der Berücksichtigung zeitlicher und räumlicher Varianz und der Eignung der verwendeten Detektorsysteme geäußert. Deshalb wurden verschiedene Methoden und Detektorsysteme verglichen und das parallele Beproben mit mehreren stationären und kalibrierten automatischen Aufnahmesystemen als die am besten geeignete Methode zur verlässlichen und vergleichbaren Fledermausaktivitäts-Erfassung befunden.
Mit dieser Methode wurden die Fledermaus-Diversitäten und Aktivitäten in verschiedenen landwirtschaftlichen Kulturen, Wiesen und Wäldern aufgenommen. Außerdem wurde gleichzeitig die Verfügbarkeit von fliegenden Insekten (potentieller Fledermausbeute) mit Licht- oder Klebefallen erfasst. In mehr als 500 Erfassungsnächten wurden circa 110,000 akustische Fledermaus-Rufsequenzen und nahezu 120,000 nachtaktive Insekten gesammelt. Insgesamt wurden 14 Fledermausarten nachgewiesen, darunter die im Gebiet sehr seltene und stark bedrohte Nordfledermaus (Eptesicus nilssonii) und die Mopsfledermaus (Barbastella barbastellum). Alle Arten wurden auch auf landwirtschaftlich genutzten Flächen detektiert.
Die Landwirtschaft im südlichen Rheinland-Pfalz ist durch Weinanbau geprägt. Die Untersuchungen zeigten, dass Weinberge aufgrund der geringen Verfügbarkeit an kleineren nachtaktiven Insekten für die meisten Fledermausarten nur eine geringe Qualität als Jagdgebiet haben. Ein weiterer paarweiser Vergleich von Weinbergen und benachbarten Regenrückhaltebecken bezüglich Nahrungsverfügbarkeit und Fledermausaktivität zeigte, dass Regenrückhaltebecken wichtige Nahrungshabitate im Weinbaugebiet darstellen. Das Anlegen dieser künstlichen Kleingewässer ist somit eine geeignete Methode um Nahrungshabitate für Fledermäuse zu schaffen und damit den negativen Effekten der konventionellen Landwirtschaft hinsichtlich der Nahrungsverfügbarkeit entgegenzuwirken. In anderen landwirtschaftlichen Kulturen wurden mit den parallel untersuchten Wald- und Wiesenhabitaten vergleichbar hohe Insektenvorkommen und Fledermausaktivitäten nachgewiesen. Besonders hohe Fledermausaktivitäten so wie eine besonders hohe Verfügbarkeit von geeigneten Nahrungstieren wurden in Apfelplantangen und Gemüsefeldern gemessen. Da diese beiden Kulturen hohem Pestizidaufwand unterliegen, kann eine Exposition von Fledermäusen gegenüber Pflanzenschutzmitteln dort nicht ausgeschlossen werden. Um das zurzeit verwendete Verfahren zur Risikoabschätzung von Pflanzenschutzmittel-anwendungen auf Vögel und Säuger auf Fledermäuse zu übertragen, wurden nach Applikation eines Insektizides dessen Rückstände auf Fledermaus-artspezifischen Beuteinsekten gemessen. Parallel dazu wurde die Fledermausaktivität erfasst. Die höchsten Pestizidrückstände wurden auf kronenbewohnenden Insekten und Spinnen nachgewiesen. Die darauf basierende Risikoabschätzung deutet auf ein Langzeitrisiko für alle Fledermausarten, die sich wenigstens zum Teil von kronenbewohnenden Arthropoden ernähren, hin. Das Vorkommen von Fledermäusen in landwirtschaftlichen Flächen, die einen Großteil der europäischen, aber auch der weltweiten Fläche ausmachen, führt je nach Kultur zur Exposition von Fledermäusen durch kontaminierte Nahrung oder zu einer Verringerung von Beuteinsekten. Bisher konzentrierten sich Schutzbemühungen auf die Sicherung von Winterquartieren und die Bereitstellung künstlicher Tagesquartiere. Potentielle Effekte von Pflanzenschutzmitteln auf Agrarflächen haben aber möglicherweise einen entscheidenden Einfluss auf die Populationsgrößen von Fledermausarten die in diesen Gebieten vorkommen. Aus diesem Grund sollten die Effekte von Pflanzenschutzmitteln auf Fledermäuse untersucht werden, insbesondere bei der Risikoabschätzung im Pflanzenschutzmittel-Zulassungsverfahren.
Invasive species often have a significant impact on the biodiversity of ecosystems and the species native to it. One of the worst invaders worldwide is Aphanomyces astaci, the causative agent of the crayfish plague, an often fatal disease to crayfish species not native to North America. Aphanomyces astaci originates from North America and was introduced to Europe in the midst of the 19th century. Since then, it spread throughout Europe diminishing the European crayfish populations. The overall aim of this thesis was to evaluate the threat that A. astaci still poses to European crayfish species more than 150 years after its introduction to Europe. In the first part of the thesis, crayfish specimens, which are available in the German pet trade, were tested for infections with A. astaci. Around 13% of the tested crayfish were clearly infected with A. astaci. The study demonstrated the potential danger the pet trade poses for biodiversity through the import of alien species and their potential pathogens, in general. In the second part of the thesis, the A. astaci infection prevalence of crayfish species in wild populations in Europe was tested. While the stone crayfish, Austropotamobius torrentium, showed high susceptibility to different haplogroups of A. astaci, the narrow-clawed crayfish, Astacus leptodactylus, was able to survive infections, even by haplogroup B, which is considered to be highly virulent. In the last part of the thesis, A. astaci was traced back to its original distribution area of North America. While the crayfish plague never had such a devastating effect on crayfish in North America as it had in Europe, the reasons for the success of invasive crayfish within North America are not yet fully understood. It is possible that A. astaci increases the invasion success of some crayfish species. Several populations of the rusty crayfish, Orconectes rusticus, in the Midwest of North America were confirmed to be infected with A. astaci and a new genotype was identified, possibly indicating that each crayfish host is vector of a unique A. astaci genotype, even in North America. Overall, the present thesis provides evidence that A. astaci is still a major threat to the crayfish species indigenous to Europe. Crayfish mass mortalities still occur in susceptible crayfish species like A. torrentium even 150 years after the first introduction of A. astaci. While there are some indications for increased resistances through processes of co-evolution, the continuous introduction of crayfish species to Europe threatens to cause new outbreaks of the crayfish plague through the parallel introduction of new, highly virulent A. astaci strains.
Diet-related effects of antimicrobials in aquatic decomposer-shredder and periphyton-grazer systems

Leaf-associated microbial decomposers as well as periphyton serve as important food sources for detritivorous and herbivorous macroinvertebrates (shredders and grazers) in streams. Shredders and grazers, in turn, provide not only collectors with food but also serve as prey for predators. Therefore, decomposer-shredder and periphyton-grazer systems (here summarized as freshwater biofilm-consumer systems) are highly important for the energy and nutrient supply in heterotrophic and autotrophic stream food webs. However, both systems can be affected by chemical stressors, amongst which antimicrobials (e.g., antibiotics, fungicides and algaecides) are of particular concern. Antimicrobials can impair shredders and grazers not only via waterborne exposure (waterborne effect pathway) but also through dietary exposure and microorganism-mediated alterations in the food quality of their diet (dietary effect pathway). Even though the relevance of the latter pathway received more attention in recent years, little is known about the mechanisms that are responsible for the observed effects in shredders and grazers. Therefore, the first objective of this thesis was to broaden the knowledge of indirect antimicrobial effects in a model shredder and grazer via the dietary pathway. Moreover, although freshwater biofilm-consumer systems are most likely exposed to antimicrobial mixtures comprised of different stressor groups, virtually nothing is known of these mixture effects in both systems. Therefore, the second objective was to assess and predict diet-related antimicrobial mixture effects in a model freshwater biofilm-consumer system. During this thesis, positive diet-related effects of a model antibiotic on the energy processing and physiology of the shredder Gammarus fossarum were observed. They were probably triggered by shifts in the leaf-associated microbial community in favor of aquatic fungi that increased the food quality of leaves for the shredder. Contrary to that, a model fungicide induced negative effects on the energy processing of G. fossarum via the dietary pathway, which can be explained by negative impacts on the microbial decomposition efficiency leading to a reduced food quality of leaf litter for gammarids. For diet-related antimicrobial effects in periphyton-grazer systems, a model algaecide altered the periphyton community composition by increasing nutritious and palatable algae. This resulted in an enhanced consumption and physiological fitness of the grazer Physella acuta. Finally, it was shown that complex horizontal interactions among leaf-associated microorganisms are involved, making diet-related antimicrobial mixture effects in the shredder G. fossarum difficult to predict. Thus, this thesis provides new insights into indirect diet-related effects of antimicrobials on shredders and grazers as well as demonstrates uncertainties of antimicrobial mixture effect predictions for freshwater biofilm-consumer systems. Moreover, the findings in this thesis are not only informative for regulatory authorities, as indirect effects and effects of mixtures across chemical classes are not considered in the environmental risk assessment of chemical substances, but also stimulate future research to close knowledge gaps identified during this work.
Agricultural land-use may lead to brief pulse exposures of pesticides in edge-of-field streams, potentially resulting in adverse effects on aquatic macrophytes, invertebrates and ecosystem functions. The higher tier risk assessment is mainly based on pond mesocosms which are not designed to mimic stream-typical conditions. Relatively little is known on exposure and effect assessment using stream mesocosms.
Thus the present thesis evaluates the appliacability of the stream mesocosms to mimic stream-typical pulse exposures, to assess resulting effects on flora and fauna and to evaluate aquatic-terrestrial food web coupling. The first objective was to mimic stream-typical pulse exposure scenarios with different durations (≤ 1 to ≥ 24 hours). These exposure scenarios established using a fluorescence tracer were the methodological basis for the effect assessment of an herbicide and an insecticide. In order to evaluate the applicability of stream mesocosms for regulatory purposes, the second objective was to assess effects on two aquatic macrophytes following a 24-h pulse exposure with the herbicide iofensulfuron-sodium (1, 3, 10 and 30 µg/L; n = 3). Growth inhibition of up to 66 and 45% was observed for the total shoot length of Myriophyllum spicatum and Elodea canadensis, respectively. Recovery of this endpoint could be demonstrated within 42 days for both macrophytes. The third objective was to assess effects on structural and functional endpoints following a 6-h pulse exposure of the pyrethroid ether etofenprox (0.05, 0.5 and 5 µg/L; n = 4). The most sensitive structural (abundance of Cloeon simile) and functional (feeding rates of Asellus aquaticus) endpoint revealed significant effects at 0.05 µg/L etofenprox. This concentration was below field-measured etofenprox concentrations and thus suggests that pulse exposures adversely affect invertebrate populations and ecosystem functions in streams. Such pollutions of streams may also result in decreased emergence of aquatic insects and potentially lead to an insect-mediated transfer of pollutants to adjacent food webs. Test systems capable to assess aquatic-terrestrial effects are not yet integrated in mesocosm approaches but might be of interest for substances with bioaccumulation potential. Here, the fourth part provides an aquatic-terrestrial model ecosystem capable to assess cross-ecosystem effects. Information on the riparian food web such as the contribution of aquatic (up to 71%) and terrestrial (up to 29%) insect prey to the diet of the riparian spider Tetragnatha extensa was assessed via stable isotope ratios (δ13C and δ15N). Thus, the present thesis provides the methodological basis to assess aquatic-terrestrial pollutant transfer and effects on the riparian food web.
Overall the results of this thesis indicate, that stream mesocosms can be used to mimic stream-typical pulse exposures of pesticides, to assess resulting effects on macrophytes and invertebrates within prospective environmental risk assessment (ERA) and to evaluate changes in riparian food webs.
Systemische Neonicotinoide gehören zu den weltweit meist genutzten Insektiziden. Neben ihrer Anwendung in der Landwirtschaft werden sie zunehmend zur Bekämpfung von Baumschädlingen in
der Forstwirtschaft eingesetzt. Die im Herbst von Laubbäumen fallenden Blätter können allerdings
immer noch Neonicotinoide enthalten. Gelangen diese kontaminierten Blätter schließlich in
nahegelegene Bäche werden die wasserlöslichen Neonicotinoide wieder mobilisiert und somit
potenziell aquatische Nicht-Zielorganismen über die Wasserphase exponiert. Obwohl der Standardtestorganismus Daphnia magna (Crustacea; Cladocera) relativ unempfindlich gegenüber
Neonicotinoiden ist, sind viele andere aquatische Invertebraten bereits bei einer Exposition im ng/L- bis niedrigem μg/L-Bereich negativ beeinträchtigt. Besonders laubzersetzende Invertebraten (= Shredder) könnten, zusätzlich zu einer Exposition über die Wasserphase, durch den Eintrag von Neonicotinoid-kontaminiertem Laub in ein Fließgewässer negativ beeinträchtigt werden, da Laub für sie eine essentielle Nahrungsquelle darstellt. Jedoch erhielt dieser Expositionspfad im Zusammenhang mit aquatischen Shreddern und Neonicotinoid-kontaminiertem Pflanzenmaterial bisher kaum Aufmerksamkeit seitens der Forschung und findet keine Berücksichtigung in der aquatischen Umweltrisikobewertung. Das Hauptziel dieser Arbeit war daher (1) Neonicotinoidrückstände in Blättern zu quantifizieren sowie für Shredder relevante Expositionswege zu identifizieren, (2)
ökotoxikologische Effekte einer Exposition über die Wasserphase sowie über die Nahrung für zwei
Modell-Shredder Gammarus fossarum (Amphipoda) und Chaetopteryx villosa (Insecta) zu untersuchen, und schließlich (3) biotische und abiotische Faktoren zu betrachten, welche eine Exposition unter Feldbedingungen potenziell beeinträchtigen könnten.
Im Rahmen dieser Arbeit konnten Rückstände der Neonicotinoide Imidacloprid, Thiacloprid und
Acetamiprid in Blätter behandelter Schwarzerlen quantifiziert werden. Ein entwickeltes „Worst-Case
Modell“ prognostizierte niedrige Imidaclopridwasserkonzentrationen für einen Bach in welchen Imidacloprid-kontaminierte Blätter eingetragen werden. Jedoch konnte mit Hilfe des Modells die Aufnahme über die Nahrung als ein für aquatische Shredder relevanter Expositionspfad identifiziert werden. Der Konsum von Neonicotinoid-kontaminierten Blättern führte, bei gleichzeitiger Exposition über die Wasserphase (= kombinierte Exposition), in beiden Testorganismen zu stärkeren Effekten als die alleinige Exposition über die Wasserphase. Des Weiteren gelang es in einem weiteren Laborexperiment die beiden Expositionswege mittels einer Durchflussanlage zu separieren. Hierbei führte die separate Exposition von G. fossarum sowohl über die Nahrung (= Konsum von Thiaclopridkontaminierten Blättern) als auch über die Wasserphase zu vergleichbaren Effektgrößen. Zudem ließen sich die unter einer kombinierten Exposition beobachteten Effektgrößen weitestgehend mit dem Referenzmodell der „Unabhängigen Wirkung“ vorhersagen, was eine Wirkung auf unterschiedliche molekulare Zielorte vermuten lässt. Die durch Imidacloprid ausgelöste toxischen Effekte auf G. fossarum konnten schließlich durch eine Behandlung der Blätter mit UV-Strahlung (repräsentativ für Sonnenlicht) sowie durch Leaching in Wasser reduziert werden. Jedoch waren beide Shredder-Spezies nicht dazu in der Lage aktiv eine Aufnahme von Neonicotinoiden über die Nahrung zu vermeiden. Daher geht aus dieser Arbeit die Empfehlung hervor, bereits während der Registrierung von systemischen Pestiziden, auf nahrungsbedingte Effekte zu testen und dadurch aquatische Shredder als auch assoziierte Ökosystemfunktionen (z.B. Laubabbau) zu schützen.
Leaf litter breakdown is a fundamental process in aquatic ecosystems, being mainly mediated by decomposer-detritivore systems that are composed of microbial decomposers and leaf-shredding, detritivorous invertebrates. The ecological integrity of these systems can, however, be disturbed, amongst others, by chemical stressors. Fungicides might pose a particular risk as they can have negative effects on the involved microbial decomposers but may also affect shredders via both waterborne toxicity and their diet; the latter by toxic effects due to dietary exposure as a result of fungicides’ accumulation on leaf material and by negatively affecting fungal leaf decomposers, on which shredders’ nutrition heavily relies. The primary aim of this thesis was therefore to provide an in-depth assessment of the ecotoxicological implications of fungicides in a model decomposer-detritivore system using a tiered experimental approach to investigate (1) waterborne toxicity in a model shredder, i.e., Gammarus fossarum, (2) structural and functional implications in leaf-associated microbial communities, and (3) the relative importance of waterborne and diet-related effects for the model shredder.
Additionally, knowledge gaps were tackled that were related to potential differences in the ecotoxicological impact of inorganic (also authorized for organic farming in large parts of the world) and organic fungicides, the mixture toxicity of these substances, the field-relevance of their effects, and the appropriateness of current environmental risk assessment (ERA).
In the course of this thesis, major differences in the effects of inorganic and organic fungicides on the model decomposer-detritivore system were uncovered; e.g., the palatability of leaves for G. fossarum was increased by inorganic fungicides but deteriorated by organic substances. Furthermore, non-additive action of fungicides was observed, rendering mixture effects of these substances hardly predictable. While the relative importance of the waterborne and diet-related effect pathway for the model shredder seems to depend on the fungicide group and the exposure concentration, it was demonstrated that neither path must be ignored due to additive action. Finally, it was shown that effects can be expected at field-relevant fungicide levels and that current ERA may provide insufficient protection for decomposer-detritivore systems. To safeguard aquatic ecosystem functioning, this thesis thus recommends including leaf-associated microbial communities and long-term feeding studies using detritus feeders in ERA testing schemes, and identifies several knowledge gaps whose filling seems mandatory to develop further reasonable refinements for fungicide ERA.
The application of pesticides to agricultural areas can result in transport to adjacent non-target environments. In particular, surface water systems are likely to receive agricultural pesticide input. When pesticides enter aquatic environments, they may pose a substantial threat to the ecological integrity of surface water systems. To minimize the risk to non-target ecosystems the European Union prescribes an ecotoxicological risk assessment within the registration procedure of pesticides, which consists of an effect and an exposure assessment.
This thesis focuses on the evaluation of the exposure assessment and the implications to the complete regulatory risk assessment, and is based on four scientific publications. The main part of the thesis focuses on evaluation of the FOCUS modelling approach, which is used in regulatory risk assessment to predict pesticide surface water concentrations. This was done by comparing measured field concentrations (MFC) of agricultural insecticides (n = 466) and fungicides (n = 417) in surface water to respective predicted environmental concentrations (PEC) calculated with FOCUS step 1 to step 4 at two different levels of field relevance. MFCs were extracted from the scientific literature and were measured in field studies conducted primarily in Europe (publications 1 and 3).
In addition, an alternative fugacity-based multimedia mass-balance model, which needs fewer input parameters and less computing effort, was used to calculate PECs for the same insecticide MFC dataset and compared to the FOCUS predictions (publication 3). Furthermore, FOCUS predictions were also conducted for veterinary pharmaceuticals in runoff from an experimental plot study, to assess the FOCUS predictions for a different class of chemicals with a different relevant entry pathway (publication 2).
In publication 4, the FOCUS step-3 approach was used to determine relevant insecticide exposure patterns. These patterns were analysed for different monitoring strategies and the implications for the environmental risk assessment (publication 4).
The outcome of this thesis showed that the FOCUS modelling approach is neither protective nor appropriate in predicting insecticide and fungicide field concentrations. Up to one third of the MFCs were underpredicted by the model calculations, which means that the actual risk might be underestimated. Furthermore, the results show that a higher degree of realism even reduces the protectiveness of model results and that the model predictions are worse for highly hydrophobic and toxic pyrethroids.
In addition, the absence of any relationship between measured and predicted concentrations questions the general model performance quality (publication 1 and 3). Further analyses revealed that deficiencies in protectiveness and predictiveness of the environmental exposure assessment might even be higher than shown in this thesis, because actual short-term peak concentrations are only detectable with an event-related sampling strategy (publication 4). However, it was shown that the PECs of a much simpler modelling approach are much more appropriate for the prediction of insecticide MFC, especially for calculations with a higher field relevance (publication 3). The FOCUS approach also failed to predict concentrations of veterinary pharmaceuticals in runoff water (publication 2). In conclusion, the findings of this thesis showed that there is an urgent need for the improvement of exposure predictions conducted in the environmental risk assessment of pesticides as a group of highly relevant environmental chemicals, to ensure that the increasing use of those chemicals does not lead to further harmful effects in aquatic ecosystems.
